Lotions, creams, medicines, etc will not effect the test. Plus, CheckMate is such a well built product that if you want to see for yourself, there is more than enough material to test everything in the house including things like lotions, creams, medicines, etc. That way you can see for yourself that all of these things do not effect the test.

What if the guy she is seeing has had a vasectomy? 

Yes, CheckMate detects semen from men who have had this procedure performed. Semen and sperm are two different things. A vasectomy does stop the delivery of sperm but ejaculation and semen production remains normal. 

I'm a woman - Does it show female semen on his underwear?

Only men produce semen and there is no reason for a guy to come home with semen stains in his underwear. Do I need a U.V or "black light" to use CheckMate?

No, a black light is not needed to perform the CheckMate test. The ultra violet light is for when you're testing a large area like a blanket or carpet and you don't know where to begin testing. A light cannot identify a stain as semen, but once you isolate a stained area with a light, then you can test it with CheckMate.

Have you seen those commercials on tv advertising black lights to locate cat urine stains? Now, do you think a black light can identify a semen stain? Of course not, thousands of different substances will fluoresce when subjected to ultra violet light, that's why some people say C.S.I. actually stands for "Crime Show Inaccuracy," because things simply do not happen like you see on television.

A black light can however be a handy thing to carry in your suitcase because it just about guarantees a hotel room upgrade every time when you scan the bedspread and discover one or two hundred different stains left by other people and their pets. Most people don't notice the fact that the maids are only changing the sheets and pillow cases, but it's true, and even though in most cases you will certainly find countless stains on any unwashed hotel bedspread, for the most part the stains will all look the same under a U.V. light and there is simply no way to tell a semen stain from coffee with cream and sugar.
